Effect of growth regulators on in vitro shoots of Gladiolus hybridus

Cormels of gladiolus cv. Cream White were cultured after descaling in MS medium supplemented with BAP and kinetin. Shoots obtained in vitro were excised aseptically and cultured in MS medium supplemented with BAP, kinetin, and NAA. Maximum culture weight and callus with axillary bud weight were obtained in MS supplemented with 1 mg/1 BAP which was found to be better than kinetin. Absence of growth substances induced long single shoots. The addition of NAA to the basal medium enhances rooting with single long shoots. BAP at 0.75 mg/l gave the maximum number (16) of shoots and axillary (19.3) buds. Correlation studies indicated that shoot number bears a direct relationship with culture weight, bud number, and bud+callus weight.
