Some characteristics of Odontoglossum ringspot virus

102 orchid plants were tested by means of ELISA for the presence of CyMV (Cymbidium mosaic virus), ORSV (Odontoglossum ringspot virus), and CyRSV (Cymbidium ringspot virus). Fifty two plants were infected with CyMV, while ORSV was found in 55 plants. Positive reactions were obtained from plants without symptoms as well as from orchids with symptoms. CyRSV was not detected in the ELISA tests. Two viruses were isolated from the plant which reacted positively in ELISA tests for CyMV and ORSV. One of them, transmitted by sap inoculation to Chenopodium quinoa, resembled ORSV. The virus induced local and systematic infection in Capsicum annuum, Nicotiana benthamiana, and N. clevelandii and local infection in 13 of 28 species tested. The dilution end point was 10-6-10-7. The thermal inactivation point was 92-94°C. The virus remained infective in Ch. quinoa sap for 3 but not 4 months. Three bands were observed when the virus was centrifuged in sucrose density gradient. The upper one consisted of short rigid particles, while the lower two contained mainly long particles. The virus showed no relation with antisera to carnation mottle and cymbidium mosaic viruses. The prepared antiserum reacted positively with the sap of infected orchids, the strains of tomato mosaic virus and ribgrass mosaic virus.

The other virus mechanically transmitted from Cymbidium with a high A490 absorption value for CyMV to Datura stramonium needs more intensive study. Typical symptoms of CyMV were induced in D. stramonium but the authors failed to transmit the virus to the other host plants.
